提供了一個scRNA-seq鑒定細胞亞型的套路
(明明我也是這樣別人卻發(fā)了Science系列):
Strategy for discovery and validation of DC and monocyte subtypes
To determine the subtypes of DCs and monocytes in human blood, we developed an experimental and computational strategy to
(i) perform single-cell RNA sequencing on DCs and monocytes derived from a single healthy individual; 單細胞測序隔盛,獲得健康組織數(shù)據(jù)
(ii) identify clusters of cells that are similar to each other; 聚類分析
(iii) find discriminative markers per cluster; 找出區(qū)分每個類別的marker基因
(iv) prospectively isolate cells corresponding to key clusters using newly identified surface markers; 利用新鑒定的marker基因將一些潛在的細胞亞型從大類中分出來
(v) validate the identity of the sorted cells using scRNA-seq;
利用scRNA-seq鑒定挑選的細胞特性
(vi) confirm the existence of these cell types in up to 10 independent healthy individuals; and 確認這些細胞類型普遍存在于多個樣品
(vii) perform functional analyses for selected cell types. 指定細胞類型的功能分析
個人認為最艱難的還是第3和第4步犹菱,不把文獻庫翻箱倒柜..把該組織和各個細胞以及它們的marker基因搞懂的話..要出好結果還是很難的....
