研究背景
動脈粥樣硬化(AS)性心血管疾病作為全球人口死亡的重大疾病之一乍桂,其發(fā)病常與營養(yǎng)膳食不均密切相關沪猴。因此图谷,人們通過攝入可降低AS發(fā)病風險的含非營養(yǎng)素成分的果蔬來防控AS的發(fā)生蚌本、發(fā)展嵌莉。
有研究證實,巨噬細胞炎癥是AS發(fā)病的重要因素蚕涤,且受NF-kB(p50尸曼、p65亞基)異二聚體調控冒签。在休眠期的巨噬細胞中,胞漿中的NF-kB與其抑制劑IkBα結合,但當巨噬細胞遭受促炎因子刺激時,IkB激酶(IKK)被激活,進而使IkBα磷酸化琳水,導致IkBα發(fā)生泛素化及蛋白酶體降解淮摔;NF-kB被釋放后五辽,NF-kB核定位序列暴露建丧,由胞漿動力蛋白Dynein將NF-kB以負極運動方向運輸至胞質蛋白importin-??5上叁熔,importin-??5護送NF-kB從胞質向核內(nèi)轉移委乌,啟動多個基因的轉錄,例如促炎因子IL-1β荣回、TNF-α的表達遭贸,那么,抑制巨噬細胞中NF-kB激活可能是治療AS的潛在途徑心软。倍半萜類化合物山萵苣苦素(Lactucopicrin壕吹,LCP)作為一種分布于菊科蔬菜生菜、菊科藥用植物菊苣中的苦味物質删铃,具抗瘧耳贬、抗癌、鎮(zhèn)痛鎮(zhèn)靜等藥理活性猎唁,但其對AS的影響未見報道咒劲。故中山大學公共衛(wèi)生院王冬亮教授團隊基于RAW264.7巨噬細胞模型和ApoE基因敲除小鼠模型探討LCP對AS的影響及功能機制。
